Dual-Gene Research Points Toward a New Approach to MSUD
Peer-reviewed research in Science Translational Medicine reports preclinical results from a dual-gene strategy designed to address the two most common forms of Maple Syrup Urine Disease.
For families living with Maple Syrup Urine Disease (MSUD), progress begins with a clear understanding of what the disease takes away and what a new therapy would need to restore. In 2025, researchers reported an important preclinical step toward that goal in Science Translational Medicine.
The study evaluated a one-time dual-gene replacement strategy designed to deliver working copies of BCKDHA and BCKDHB, the two genes associated with the most common forms of MSUD. In mouse models and a naturally occurring calf model, the approach restored key metabolic function and supported survival and normal dietary tolerance. These findings are preclinical, which means the therapy has not yet been studied in people, but they provide important evidence for continued development.
Plowshare’s lead program builds on this work as we advance the careful scientific, manufacturing, toxicology, and regulatory steps required before a potential clinical trial.
